I don't know about "defense", but I'll at least offer some commentary.
Mai Tokiha: Youko Honna. Based on her performance as Nagisa/Cure Black from Pretty Cure, she has the range, and I just like her voice.
Mikoto Minagi: Miina Tominaga. I admit having trouble with the required characters, especially Mikoto. Then as I was watching Patlabor I realized Noa Izumi's voice could handle both the cute focused nature of the character, and the intensity that shows up later.
Natsuki Kuga: Nana Mizuki. If Mai-HiME has a Fate Testarossa character (or Utau Hoshina, or Rue of course), surely it's Natsuki. Unless it's Nao. But Nao's hair is too short. In any case, Nana is well suited to the standoffish nature of the character, as well as her moments when that reserve breaks down.
Yuuichi Tate: Kousuke Okano. Thinking of Recca, or Asuka Jr. More or less heroic, but a little out of his depth.
Nagi Homura: Noriko Hidaka. Specifically I was thinking of Sojiro Seta of Rurouni Kenshin, for a depiction of an obnoxious yet threatening kid. She was also Jean from Nadia, for, well, not quite the same sort of character.
Reito Kanzaki: Takeshi Kusao. Sexy voice with a touch of arrogance. Take his Saionji from Utena, and dial it down slightly. Or Krad works as well.
Nao Yuuki: Chiyako Shibahara. Nobody knows her, but I love her, so I picked her. Her big role was 8 1/2 tails foxgirl Sakura from Hyper Police. I also like her as the idol robot operator who bailed on everyone in Key the Metal Idol. A sexy, predatory growl of a voice, well suited to Nao.
Miyu Greer: Megumi Toyoguchi. I want a frighteningly cold Miyu, so I'm thinking of Revy in her calmer, decidedly-unimpressed-with-Rock moments here.
Yukino Kikukawa: Halko Momoi. Nurse Witch Komugi in her more put-upon moments should be able to voice constantly-put-upon Yukino.
Haruka Suzushiro: Yumi Takada. Haruka is more or less an Ojou-sama in training, so I thought it would be good to cast a classic Ojou-sama for her. For all of her roles, Ayeka remains preeminent.
Shizuru Fujino: Sayaka Ohara. Think of Yuuko from XXXHolic for a seductive voice, with secrets.
Akira Okuzaki: Yu Kobayashi. Hearing Setsuna from Negima reminded me that that sort of tomboyish voice would work well for Akira.

Mai Tokiha: Yuko Minaguchi
Yuko Minaguchi's voice acting as the kind, honorable, brave, & yet romantically insecure Yawara!, and her voice tone which changed subtly as Yawara grew older, suit Mai Tohika's character very well.
Mikoto Minagi: Maya Okamoto
Maya Okamoto has voiced adults such as Emma Sheen in Z Gundam, & spunky younger sister characters like Reina in ZZ Gundam. She can voice determined, fierce & tender girls excellently.
Natsuki Kuga: Rei Sakuma
Rei Sakuma has voiced older/college girls & adult women in many action shows. Kazumi in Gunbuster, Peorth in Ah! My Goddess, Aika in Agent Aika & Shampoo in Ranma. For a more mature girl than her contemporaries, Rei Sakuma would perform Natsuki perfectly.
Yuuichi Tate: Tohru Furya
Tohru Furya's experience playing teens & young adults is immense. Whether slightly awkward guys like Kyosuke Kasaga in Kimagure Orange Road, or heroes such as Amauro in MS Gundam. Yuuichi Tate's character is one he can handle with ease.
Nagi Homura: Mamoru Miyano
Mamoru Miyano is the voice of Light in Death Note. His skill at playing Light, the scheme, self-made Punisher with a pen in Death Note, qualifies him as the ideal voice to play the amoral Nagi in My-Hime, for whom norman humans are expendable.
Reito Kanzaki: Masanori Ikeda
Masanori Ikeda's performance voicing Shishio Makoto in Rurouni Kenshin, a villain with a sense of honour & his own code, combined with the deeper voice Reito Kanzaki needs, is an ideal casting of character-voice actor with a more mature student who has an air of mystery about him.
Takumi Tohika: Akira Ishida (Optional 1)
Cast with a deliberate irony as Mai's younger, delicate but brave brother. This is a part Akira Ishida can voice well as his Chrono in Chrono Crusade is a moving performance.
Shizuru Fujino: Hiromi Tsuru (Optional 2)
Hiromi Tsuru has recently been voicing adult roles, but her ability includes younger girls & more mature mid-teens onward. She has also voiced sexy femme fatales such as Madam Midorikawa in Great Dangaioh & Camille in Master of Mosquiton. Shizuru in My-Hime, is a character that could have been written especially for Hiromi Tsuru, as Shizuru's personality opens up in the story.
Haruka Suzushiro: Mitsuko Saiga (Optional 3)
Mitsuko Saiga has voiced many well know & appreciated female characters with strong personalities & sometimes abrasive manners. These include Sobaash from Banner of the Stars, Countess from Gilgamesh & Makoto from Genshiken. Voicing Haruka in My-Hime, she would give a strong & memorable performance as the brash Executive Director of the Student Council.
Midori Sugiura: Kotono Mitsuishi (Optional 4)
Kotono Mitsuishi should need no introduction. She is one of the best known voice actresses in Japan, & globally. Kotono has voiced cute characters, kids, teens & adults. Kotono's latest (reprised) adult character is Misato Katsuragi in the first rebuild Evangelion movie "You Are (Not) Alone". Midori Sugiura is a lighter role, but similar, in that the character acts as though she lives on the light side, but has a more serious, but still high adrenalin side & purpose to her character.
Akane Higurashi: Yuri Shiratori (Optional 5)
Yuri Shiratori charms in the gentle characters she voice acts, such as Tsubame in Rurouni Kenshin, Angelique in the shows of that name, & Sayo in Negima.
Yukariko Sanada: Akiko Hiramatsu (Optional 6)
The seiyuu needed for Yukariko Sanada, My Hime's tormented yet compassionate nun, is Akiko Hiramatsu. Akiko's experience covers characters such as Miyuki in You're Under Arrest - a sympathetic but very devoted policewoman, Bloodberry from the Saber Marionette series, and Edel in Princess Tutu.

It took some time to finalize the cast. I had trouble with figuring out what role to give X. That is until I realized I already had an idea in mind. I just needed to give Y a different role.
We'll see if I'm able to give any form of cast defense later. Also, I've told myself to accept this final decision. Hopefully I'll listen to me.

Well it's not so much a defense but rather my thought process as I went about doing my casting and a bit of commentary.
Mai Tokiha: Kanae Itou
To be honest, it was hard to chose between the 2 Kanae I had drafted up to play Mai, but from what I've heard from her part as Amu from Shugo Chara and Koume from Taisho Yakyuu Musume, she edge out Kanae Oki a bit.
Mikoto Minagi: Rie Kugimiya
Why not? Seriously if this is not type-casting, I don't know what is.
Natsuki Kuga: Yuka Komatsu
From the only show that I've heard her on where she plays a bad-ass villain who turns good, there is that certain spark and tone in her that can handle this role quite comfortably.
Yuuichi Tate: Daisuke Ono
Well, this too was a no brainer as Daisuke can pull off any young male leads with ease.
Nagi Homura: Kappei Yamaguchi
Rokuta/Enki from the Twelve Kingdom is what I had in mind when I cast him.
Reito Kanzaki: Kouichi Yamadera
One word: ALBEDO! Hence this is why the guys on my draft aren't that inspiring.
Nao Yuuki: Ryoka Yuzuki
Cool girl gone nasty? It just mirrors her performance in Buso Renkin as Tokiko Tsumura.
Miyu Greer: Kikuko Inoue
Most don't usually give thought about her mid-range voice that hardly befits her type cast of a) 17 year olds or b) deep sexy, but it was rather easy for me to see her playing Miyu using that range.
Alyssa Searrs: Ayahi Takagaki
Mainly for her singing voice with the combination of her role as Noe Isurugi in true tears makes for an easy choice.
Haruka Suzushiro: Kanae Oki
After assigning one of the 2 Kanaes I have, choosing a role for her was rather hard. She has the spunk so I though, why not give her the role of Haruka?
Shizuru Fujino: Hitomi Nabatame
No brainer on this one I had her pin for the role. You can put it up with the rest of her roles that involves yuri loving student council presidents.
Shiho Munakata: Ai Shimizu
I don't exactly remember the real reasons other than "Yes! Easy replacement for Sakura Nogawa!"
I'm rather happy about this, but at times I'm rather conflicted about my choice with Mai and the 2 Kanaes. However there's no turning back, so I'll have to live with it.

To be honest, I'm not sure if these actually qualify as defenses. However this should give a good idea as to the warped thought process that ultimately lead me to drafting each person and casting them for their finalized roles. Also to give an idea of just how poor my explanation skills and the ability to put my thoughts together can be.
Mai Tokiha: Yuko Sanpei
My main influence for picking Yuko Sanpei was her role as Nozomi (Cure Dream) in Yes Pretty Cure 5! Listening to her speak as Cure Dream, I was sure she'd work as Mai Tokiha
Mikoto Minagi: Aki Toyosaki
The main influence here was her role as Yui of K-ON! I felt that because of the she could be good for the none serious parts of Mikoto. That, and it would be interesting to hear how Aki Toyosaki handled the heavier story aspects of Mikoto.
Natsuki Kuga: Emiri Katou
For whatever reason the idea of Kagami of Lucky Star as Natsuki greatly amused me. As such I just had to draft Emiri Katou for this purpose. I have definite confidence that Emiri can handle the role.
Yuuichi Tate: Hirofumi Nojima
While looking for male names to draft, I was reminded of the fact Hirofumi played Akatsuki in Aria. I then thought "Well, both Yuuichi and Akatsuki are capable of being confrontational sometimes". Also, his role as Yusaka in Toradora gives me indication he can have the right range for Yuuichi.
Nagi Homura: Nobuhiko Okamoto
His role as Accelerator in "A Certain Magical Index" is the main reason I ultimately chose Nobuhiko Okamoto for the role of Nagi. Sure Nagi is a bit more composed than Accelerator, but have a cruel side to them.
Reito Kanzaki: Tomokazu Sugita
Sure, the two main roles I'm familiar with for Tomokazu Sugita involving him being a bit on the sarcastic side. Yet for some reason, I just felt it would be interesting to see how he handled a role like Reito.
Midori Sugiura: Satomi Satou
My main influence for choose Satomi Satou was her role as Ritsu Tainaka in K-ON! I felt that the type of drive and energy that Ritsu has shown would work well for Midori.
Nao Yuuki: Yuko Goto
Yuko Goto is one of my favorite seiyuu so I just had to draft her as soon as possible. It's probably no surprise that she was my first choice. I ultimately settled on the idea of her as Nao Yuuki because I felt her voice would work well with that side of Nao.
Alyssa Searrs: Kana Hanazawa
The main influence here would be Zange of Kannagi. A role that shows that Kana Hanazawa is capable of being elegant sounding. As such I felt she could handle Alyssa Searrs as well.
Haruka Suzushiro: Yukari Tamura
Part of the reason I chose Yukari Tamura was because of her range. As such she'd be pretty good for filling in that final role.
Mashiro Kazahana: Kana Asumi
Her role as Yuno in Sunshine Sketch was my main influence for choosing Kana Asumi. That, and there's her role as Marin in Sea Story. I figure her voice could work well for Mashiro. Also, it would be interesting to see or rather hear how she'd handle certain scenes Mashiro has later in the series.
Shiho Munakata: Eri Sendai
The main influence for choosing Eri Sendai was her role as Milky Rose in Yes Pretty Cure 5 Go Go! After listening to her several times (mainly because I like Milky Rose a lot), I felt she'd make a good Shiho. That and it gives me a Cure Dream/Milky Rose combo of sorts.
Random Side Note
Through out the draft, I kept thinking "Darn it, I should have drafted her when I had the chance". Yet the more I think about it the more I realize that my failure might be for the best. As it's partially because I failed to draft those people that I had room to to cast the people that I did.

All of the cool kids have posted their defense, and a few others have as well, so here's mine:
Mai Tokiha: Mikako Takahashi
Obviously Ayumu was why I drafted her, but Mikako has a good deal of experience and range. If people have been watching Shangri-La this season, then this should be an obvious choice, if not - get to Crunchyroll and watch episode one. It's available free of charge and legal! Mai has a 'normal but spunky' voice, which is why her role as Kuniko fits so perfectly.
Mikoto Minagi: Kaori Fukuhara
Also inspired by Crunchyroll, this time The Koromo, of Saki fame. Now Mikoto is less evil spirited and more mischevious, so combine The Koromo with The Tsukasa (Lucky Star) and you have a fun and interesting combination that can cope with the more interesting aspects of Mikoto's character later in the series.
Natsuki Kuga: Shizuka Itou
Natsuki is a cool headed and calm person, but when angered she is a real beast. When embarrassed, she is really REALLY cute. Does this remind you of Hinagiku Katsura? If not, then you aren't doing it proporly.
Yuuichi Tate: Kisho Taniyama
Yuuichi is your standard anime guy character... Kisho's experience in several series will win him here.
Nagi Homura: Nozomu Sasaki
Eriol Hiiragizawa is a very creepy name for a supposedly english character to have, and the character too was creepy. They share a lot in common, Eriol and Nagi.
Reito Kanzaki: Jun Fukuyama
Nobody voices a calm, calculating, gentleman type better than Jun Fukuyama.
Midori Sugiura: Asami Sanada
Mad teacher? Just watch K-On!
Shizuru Fujino: Yukana
Soft, calculating, and a bit twisted... Yukana's interesting vocal quirks could also come in handy here.
Mashiro Kazahana: Haruna Ikezawa
Looking at her Yoshino of Maria Watches Over Us, we have a soft and polite voiced person capable of adding not only a bit of mystery but also some flare without getting overly angry.
Shiho Munakata: Hiromi Konno
A cute voice capable of flying off the handle with love? I can thank my Lucky Star's that I got her.
Akira Okuzaki: Saeko Chiba
A deep voice fits well here, despite the young character. Think Nagihiko from Shugo Chara.
Takumi Tokiha: Ryoko Shiraishi
And a more 'boyish' voice here for Takumi... Ryoko Shiraishi is a great choice!

Okay here's the defense.
Mai Tokiha: Ami Koshimizu: What a talent, has the range to be regular Mai and excel as Mai when she's fighting, can easily cover every emotion Mai goes through in the series. Thinking of something a tad different to her Kallen from Code Geass.
Mikoto Minagi: Ai Nonaka: Well she'll get the cuteness parts down no worry and I'm sure she can voice the fighting parts well enough.
Natsuki Kuga: Rie Tanaka: Well I think the obvious choice was to cast Rie as Nao since she does stellar job with characters like that. Thought that was too cliche and decided to cast her as Natsuki. She's got the voice and acting range to sound tough like Natsuki does usually as well as act embarrassed.
Yuuichi Tate: Daisuke Namikawa: I was thinking his voice as Rock from Black Lagoon though slightly younger would be perfect for this role. I intended to draft Miyo Irino but in looking for a replacement I couldn't believe I overlooked him initially.
Nagi Homura: Hiroshi Kamiya: He can definitely get the erratic moments of Nagi right and thanks to Tiera from Gundam 00 he can sound menacing when needed.
Reito Kanzaki: Junichi Suwabe: Suwabe has played great villain after great villain, his resume speaks for itself in that regard, I wouldn't pick another VA over him for this role. Has the calmness and charm to voice Reito as he is normally and the menace and insanity to voice him as he truly is.
Midori Sugiura: Satsuki Yukino: When I think Midori I think of genki and gar from an older voice and I felt Satsuki Yukino fits that too a tee. Something like Mion would be ideal.
Nao Yuuki: Aya Hirano: I was thinking of casting her as Mai or even Natsuki but I felt like taking a risk. I definitely think Aya has the talent and skill to do a good Nao.. but we'll never know since she never plays these roles.. at least until the prequel to Bakemonogatari gets animated :p Still when Fukuyama was casted as Lelouch people thought he was miscast.. and look at him now...
Akane Higurashi: Saori Hayami: This up and coming seiyuu would excel in the role of Akane. Her role in Seikerei shows she can sound tough when needed to as well.
Miyu Greer: Akeno Watanabe: I was thinking Akeno would use a similar voice to her role in Negima since that's slightly robotic/mechanical in voicing as well.
Shizuru Fujino: Romi Paku: I think Romi would do a top job voicing Shizuru, I think she's the best VA out of the 12 I selected. She should be right handling a mature role like Shizuru.
Mashiro Kazahana: Mai Nakahara: I'll have her use a similar voice to her Felli Loss from Chrome Shelled Regios, perfect for the role of Mashiro and sounds nothing like her Mai from the original cast.

Mai Tokiha: Kana Ueda
Kana Ueda has good range of roles and could have cast her in other roles, but I felt she was a good choice for Mai. I think she has the kind of cute and cheerful but still strong voice that fits with what I think of Mai.
Mikoto Minagi: Noriko Shitaya
Noriko Shitaya has a great voice for high-pitched cutesy characters, so she seemed like a nice fit for Mikoto. She's maybe known for demure types more (so a bit of a risk I guess), but she's done more energetic characters like Milcatopy in Kyouran Kazoku Nikki well too. Having her paired up with Kana Ueda is interesting to me for some reason as well.
Natsuki Kuga: Houko Kuwashima
This is the first role I was certain on casting. Whenever I though of Natsuki, I had to have Houko Kuwashima for her. I just love her for the cool, serious kind of girl characters. Excellent range for all aspects of her character, too.
Yuuichi Tate: Yuuichi Nakamura
Yuuichi Nakamura good for a bumbling, average kind of guy here. Even though I wasn't really thinking of it at first for some reason - Yuuichi as Yuuichi (Hah, kind of like the original cast has a Mai as a Mai, and his Clannad co-star, no less)
Nagi Homura: Daisuke Sakaguchi
Sakaguchi has good range for the younger sounding characters and pulls them off really well. I think he fits with the menacing, playful personality pretty well thinking of some of his roles like Quiche in Tokyo Mew Mew.
Reito Kanzaki: Hikaru Midorikawa
Hikaru Midorikawa commonly plays the cool, handsome types, and given what I remember of Reito, seems like he would fit like a glove.
Midori Sugiura: Natsuko Kuwatani
At first, I really wasn't sure where I was casting Kuwatani (she's a favorite of mine and thought of more of a wild card), but the more I thought, though, she's really a perfect fit for Midori. She's played some rather eccentric types ranging from cute to mature, and seemed to line up with what I'm thinking of with Midori's character.
Yukino Kikukawa: Yukari Fukui
Fukui has a nice cute, sweet voice for shy characters, like with Cocoloo in Yucie. Seems like she'd be an enjoyable take on Yukino.
Haruka Suzushiro: Akemi Kanda
I know Kanda for doing combative, exciteable characters quite a bit. She's got a prideful streak with roles like Lulu from Shugo Chara and Anteaterina in Princess Tutu, as well as the tsundere like Asuna and lead girl in Tonagura.
Shizuru Fujino: Yumi Touma
I was wanting to cast Fujino but not too entirely certain on who to go with. Yumi Touma works well because she does deeper and elegant nicely. She's got a pretty wide range of roles, making me think should could do different aspects of this character justice, too...
Akira Okuzaki: Reiko Takagi
I know Reiko Takagi for more androgynous characters and rather like her voice at that, so I though it would make for a good Akira.
Takumi Tokiha: Motoki Takagi
Motoki Takagi is absolutely perfect for Takumi in my mind. He tends to voice the young, lighter-voiced male characters excellently, which goes with the frail Takumi's personality. I was wanting to have a good number of male seiyuu even with the girl-dominated cast, so worked out well that I had a good option here.

See above. I picked her because of her Ga-Rei Zero role and so I could pair her up with Kaoru Mizuhara. She would have to use a different voice to fit Mikoto's childlike nature, but between her Coppa (Tower of Druaga) and Chiaki (Minami-ke) voices, she's clearly capable of those kinds of voices.

Marina Inoue is great for those flustered/embarrassed scenes that Natsuki gets on several occasions. I didn't have a particular role in mind when I cast her as Natsuki. Natsuki tends to be a bit colder than most of Marina Inoue's roles, that shouldn't be a problem for her. My main motivation for putting her in this role was to have her paired up with Shizuru's seiyuu, particularly for certain late series scenes.

Hiro Shimono is Mr. Male Lead, so he's overly qualified for this role. I didn't really put much thought into this one, since most people hate Tate anyway and while I don't dislike him, he's just kind of there for me and I just needed someone to fill the role. I considered casting Norio Wakamoto in this role for shits and giggles, but I couldn't justify the characterization, and since I'm otherwise playing this casting straight, it wouldn't work.

Nagi is, uh, not normal and quite a bit older than he appears, so I figured having a much older voice wouldn't be out of character and would make his scenes more amusing.

I was thinking of his role as Kuze in ef ~ a tale of memories/melodies. That exact voice seems like a good fit for Reito, especially for later in the series, since Reito's voice had that smooth and suave, almost predatory quality to it.

I drafted Masayo Kurata for her role as the batshit insane and insanely hot Fasalina in Gun x Sword and that was the only role of hers that I really new. Her character in that series was a bit older so she would have to make her voice sound a bit younger (though not by much) but otherwise, it would be a good fit for Nao.

While Sayuri Yahagi is to me Izumi Segawa first and foremost, the reason I put her in this role is her role as Rima Mashiro in Shugo Chara. Also, I wanted to hear a random "Nihahaha" while she's trying to kill people.

Yuuka Nanri originally played a bad girl role as Nao, so I decided to cast her as the completely opposite kind of character, the shy, softspoken Yukino. I was looking at her role as Karen Ichijou (School Rumble). Her Karen Ichijou voice seems perfect for Yukino.

She was actually recommended to me when I returned from vacation to find my seiyuu list destroyed and had no idea who to cast, though I cast her in a completely different type of role than the one that what was recommended to me. She has extensive experience drafting young boys, so I considered having her voice an evil young boy but ultimately decided against it. I decided that her experience as Cornelia (Code Geass) and Ayaka (various Negima anime) made her a good fit for this character. This also allowed me to cast everyone for a certain late series scene involving Natsuki, Shizuru, Haruka, and Yukino.
Shizuru Fujino: Mamiko Noto (http://www.animenewsnetwork.com/encyclopedia/people.php?id=3166)
When I drafted Mamiko Noto, I had three possible roles for her. One was a very Mamiko Noto role, but I wanted to avoid that unless I had no other choice. I wanted to hear her to a more evil or bad girl role, so I had to try to decide between casting her as Shizuru or Nao. I ended up going with Shizuru because I can here her voice coming out of Shizuru much easier than I can hear it coming out of Nao, purely from a character design perspective. In addition, this allowed me to have her and Marina Inoue play certain scenes together, which is like sex for my ears.

Kaori Nazuka played a soft-spoken, high class, wheelchair bound little girl in Code Geass, so this seemed like a perfect fit for her.

Mai Tokiha - Ryou Hirohashi
Mostly influenced by Sora Naegino from Kaleido Star combining the tough times Mai has and gets combined with her subtle and silly moments. Hints of Kyou Fujibashi from Clannad with her stubborn and tough nature also influenced this pick.
Mikoto Minagi - Kaori Mizuhashi
To pair up against Mai, we have Mikoto - and the mentality was the Rosetta Passel dynamic with Sora in similar way especially Mikoto's glomping nature. That innocent yet tough side of her fits Rosetta and Mikoto to a tee and Mizuhashi was my first pick for Mikoto.
Natsuki Kuga - Yu Asakawa
A combination of the shy, nervous Sakaki from Azumanga Daioh with the tough, no holds barred Priss Asagiri from BGC2040 was the idea of Miss Asakawa being drafted as the tough, but easily embarrassed Natsuki.
Yuuichi Tate - Noriaka Sugiyama
Weirdly, it was England from Axis Hetalia Powers which was the key for my intentional one male seiyuu. However, his nice guy mentally also fits Shiro from Fate/Stay Night with England's idiotic moments hence the pick.
Nagi Homura - Rina Satou
Nagi? Negi? That works. Besides, I've got recent chapters in the manga in the mind considering the cute, but dark nature Nagi seems to precide...
Reito Kanzaki - Megumi Ogata
Megumi seems to be the go to girl for a lot of the old school male roles, but it's a female one which people think is male was the main influence being Sailor Uranus from Sailor Moon. Tough, kind, calming but always ready for action, Ogata's calm masculine voice was the intentional pick here.
Midori Sugiura - Aya Hisakawa
My favourite seiyuu, a true veteran who can plays middle-aged women to young goddesses, this works for a 20s doctor trying to think she is much younger than she is...
Alyssa Sears - Sakura Tange
O.K, I confess. After reading Alyssa's description, my idea was 'O.K, an evvvilll Sakura Kinomoto. That works. Discussion end of. :P
Akane Higurashi - Atsuko Enomoto
The love struck Yukino Miyazawa from Kare Kano combined with the innocence of Misaki Suzuhara from Angelic Layer was the ideology here.
Haruka Suzushiro - Chinami Nishimura
Combined Mia Guillem's hard working nature with the attempts to get a rival to notice her a la Greta from Tiny Snow Fair Sugar and we have Haruka.
Shizura Fujino - Miki Ito
Oh yes, if a certain Miss Sachiko Osagawara was just the tiniest bit more evil with that low voice of hers, I'd picture Shizura. The whole shoujo-ai aspect just makes this pick even more fun.
Chie Harada - Ayana Sasagawa
She's only done one major role, but as that role is Kazumi from Negima, who else better to get as a nosey photographer? :)

Mai Tokiha: Minako Kotobuki
Minako Kotobuki has rapidly shown herself to be the most versatile of the quartet of relative newcomers to come out of K-ON!, from the Ojou-sama Tsumugi, to the evil and excitable Kanon to the Yuki-esque Kaho. Her versatility and rising star led me to put her here as the lead.
Mikoto Minagi: Momoko Saito
She’s made something of a career of playing girls who are slightly…off. From Solty in Solty Rei to Stealth Momo, she would be perfect as the breast loving, slightly feral Mikoto.
Natsuki Kuga: Youko Hikasa
Because, well….Mio. Obviously. Mio is Natsuki without the trauma and the super powers. NATSUKI CRISIS!
Yuuichi Tate: Takuma Terashima
He’s got a very “male lead” sort of voice, but is capable of being pretty awesome when he needs to be. Pretty much this would be like how he plays Teppei in Princess Lover
Nagi Homura: Minoru Shiraishi
I have no real reasoning here other than I think it would be interesting. Shiraishi’s kinda a joke, but he always does well at whatever role he’s set in, so I think it could work
Reito Kanzaki: Takehito Koyasu

Midori Sugiura: Emi Shinohara
Emi Shinohara does a lot of “Adult” roles (adult compared to the rest of the cast) and would bring a slightly different tone to Midori, but one that would work just as well.
Nao Yuuki: Haruka Tomatsu
Because I don’t think she’s played a psychopath yet. I’m positive that it would be absolutely amazing.
Miyu Greer: Kazuma Okushima
From the very serious Meiya, to the really, REALLY serious Miyu.
Shizuru Fujino: Miyuki Sawashiro
She’s recently demonstrated that she can do batshit crazy better than almost anyone else, and this is absolutely required for Shizuru.
Mashiro Kazahana: Saori Goto
She specializes in soft and sweet, but can put some iron into it when necessary, exactly what you need for Mashiro.
Shiho Munakata: Ayana Taketatsu
Imagine a bratty Azunyan.
