Well, this is very well done... loved especially the background textures though I feel the mapped texture of the closer building (the one Spidey is climbing) is a little too big for the perspective you are using... I mean... the bricks look bigger than they should be. I like the lighting on the background a lot too... but I feel it has to do a lot with the texture you chose. Nice rendering there.
Now with the characters... I have to say this... I really don't like Poser at all... it makes such lifeless models, but I have seen some that really blow me away... but those are very rare... and involve many many days of work at texturing and modeling. About your models... well...they are pretty well done... especially the clothing and accesories... you did try to give them those characteristics that make them unique... I realize that. But there is something... perhaps the fact that as much as I try to imagine this as a cinematic scene I just end looking at a maquette with action figures in pose. Poser tends to do that. Other problem is that the characters just seem to be aware of the character they are interacting with.... I mean there are a lot of characters here but they are hardly making a whole action scene. I just see Cyclops vs. The Lizard, Sue vs. Dr. Octopus and so on, but their actions (and I think this is impossible) are not altering the other characters at all.
Spiderman is like... floating over there, and seems to be somewhat small... maybe if you give him some shadow... Love the pose there... and the Daredevil pose too. The others look too static for my taste... especially Storm... You'd think her cape should be waving but she is too stiff.
There is this book... How to Draw Comics The Marvel Way, where Stan Lee says that if you want to draw a Superhero you have to exaggerate the pose, the perspective and the body proportions... with the last you succeed, but maybe you have to work more in the poses and take more dramatic angles.

Very Good work. Nice to see an 'active' group pic.
Mysterio too skinny? Daredevil too fire engine red? Kingpin too far back by the door? Mysterio and Storm have the same pose?
Lost in the shuffle: The Green Goblin. It's very hard to tell, until you see the pink mark under the feet and the two, nearly invisible diagonal lines. Like DD, there is lack of depth in the costume. Put a bit of smoke around him, in a 'C' 'S' or 'J' pattern to make the glider stand out-and so folks know who the heck he is supposed to be. (GG is in the center for those wonder who the guy in green is), otherwise, it looks like a poor attempt to throw Scorpion in there as an afterthought.
Fave characters of the piece: Doc Ock, Lizard, Spidey and Sue Storm.
* Since The Lizard, Gobby and Kingpin were never part of the Sinister Six, why call the piece Sinister Six when on the art, you name it something different (and more accurate?)
